  • New v6.0 DNA and RNA basecalling, modified base, and Dorado polish models (see Release Note for more information)
  • New Dorado smallvar tool replacing Dorado variant for SNP and INDEL calling
  • Dorado polish now provides a --model-override option and processes only the draft sequences from the input BAM file header
  • There is now a tm:Z tag in output file RG lines to indicate trimming conditions (see the Dorado Documentation SAM Specification for more information)
  • Dorado aligner now provides the -eqx minimap2 option
  • Dorado prevents excessive memory usage when running alignment
  • Dorado demux and aligner have better handling of inconsistent read group data
  • Custom barcoding now provides initial support for arrangements with dual barcodes
  • Dorado demux no longer supports consuming stdin
  • Dorado demux no longer crashes when running on previously demultiplexed files or when processing previously unclassified read groups with --no-trim flag
  • There have been improvements to reduce over-splitting of RNA reads
  • Dorado summary is now faster and the summary file output now better matches results from Dorado basecall server and the SAM specification
  • Dorado basecaller no longer crashes when there is no sample_id in the POD5 file
  • Output folder paths are fixed to include experiment ID when processing POD5s that were converted from .fast5
  • The minimum supported CUDA version has increased to 12.8 (or 12.6 on Linux ARM)
  • OpenSSL has been updated to 3.5.5
  • Pod5 has been updated to 0.3.39
  • Dorado correct now defaults to CPU mode on macOS, but allows users to specify --device metal
  • There are now pre-computed batch size benchmarks for RTX PRO 6000 Workstation cards
