オックスフォード・ナノポアテクノロジー社のユーザーグループ・ミーティング "What you're missing matters (WYMM) 2026" を東京で開催いたします。
今年は「ナノポアが拓く次世代のマルチオミクス解析」というテーマで、オックスフォード・ナノポアシーケンスの様々な活用例を、第一線でご活躍の研究者からご講演いただきます。
ナノポアの技術で、構造変異やメチル化を包括的に把握し、ずっと取り組みたいと考えていた大胆な研究課題にチャレンジし、ヒトゲノムやがんゲノムの未知の答えを一緒に探り出しましょう。希少疾患のヒトゲノムからがん研究のシーケンシングなど、最新のご講演のほか、製品展示、ナノポアコミュニティーと交流できる意見交換会、ナノポア専門家と相談する機会などが含まれます。
本イベントの参加費用は無料ですが、事前登録が必要で座席数に限りがあります。昼食と軽食が提供されます。対面式のイベントです。

Human genetic diversity cannot be fully captured by a single linear reference genome, particularly in complex genomic regions enriched for structural variation. The Japan Pangenome Project aims to establish a comprehensive representation of genomic diversity in the Japanese population using long-read sequencing and telomere-to-telomere (T2T) genome assembly. By resolving genomic regions that have remained inaccessible to conventional short-read sequencing, we are uncovering previously uncharacterized sequences, structural variants, and highly polymorphic loci that contribute to human diversity. In this talk, I will present our ongoing efforts to construct high-quality Japanese pangenome resources and expand them toward population-scale T2T genome analysis. I will highlight how long-read sequencing, including Oxford Nanopore Technologies, enables the characterization of complex genomic variation beyond the current reference genome framework. Finally, I will discuss our future vision for translating pangenome and T2T genomics into clinical research. Integrating population-specific pangenome references with large-scale disease genome datasets may improve variant discovery and interpretation, particularly in medically important but technically challenging regions, and ultimately contribute to more comprehensive genomic diagnosis and precision medicine.

河合 洋介, 国立遺伝学研究所エクソームシーケンシングによる希少疾患の診断率は、対象疾患や症例選択基準によって異なるものの、約8~50%に達することが報告されている。さらに、ショートリード全ゲノムシーケンシング、最新の知見に基づく再解析、RNAシーケンシングとの組み合わせにより、診断率を向上させることが可能である。しかし、それでもなお相当数の患者で遺伝学的原因は未解明のままであり、従来の解析では検出が困難な病的バリアントが存在することが示唆される。ロングリードシーケンシングは、ショートリード技術と比較して、構造バリアントやリピート伸長など特定のタイプのバリアントの検出、反復配列やGCリッチ領域など解析困難なゲノム領域の解析、リードに基づくバリアントのフェージング、さらに追加実験を必要とせずDNAメチル化と遺伝的バリアントを同時に解析できることなど、複数の利点を有する。実際にこれまでの研究から、ロングリードシーケンシングによって従来見逃されていた遺伝的バリアントを同定できることが示されている一方、希少疾患診断におけるその有用性については、いまだ十分なコンセンサスが得られていない。本セッションでは、ロングリードシーケンシングを用いた疾患関連構造バリアント検出の経験を紹介するとともに、希少疾患コホートの解析から得られた知見について議論する。

We prospectively evaluated targeted adaptive sampling long-read sequencing (TAS-LRS) as a rapid genomic profiling platform for pediatric hematologic malignancies in the nationwide JCCG-WGS study.
Patients with hematologic malignancies were enrolled from participating institutions of the Japan Children’s Cancer Group. TAS-LRS was performed using nanopore sequencing, targeting genes associated with hematologic malignancies and their flanking regions. The analytical workflow simultaneously assessed small variants, SVs, CNVs, and methylation-based classifications.
TAS-LRS provided clinically relevant genomic information within a short turnaround time. It identified canonical fusion genes as well as complex noncoding rearrangements consistent with enhancer hijacking, including IGH::MYC, IGH::CEBPA, and TLX3::BCL11B. Subtype-defining CNVs, including iAMP21 and near-haploidy, were also detected. In one case, identification of CBFA2T3::GLIS2 led to revision of the initial diagnosis. Methylation-based classification further supported genomic subtype assignment in a substantial proportion of evaluable cases.
These findings demonstrate the feasibility of TAS-LRS for rapid and comprehensive genomic profiling in a prospective multicenter setting and support its potential integration into real-time diagnostic and therapeutic decision-making.
