- In the section 'Available kits', timings in the images for the sequencing workflow were updated following an increase in the flow cell load timing from 5 min to 10 min.
- The Tables for DNA quantification methods in sections 3, 'Ligation-based sequencing kits', and 4, 'Rapid-based sequencing kits' were moved to section 8, 'Sample input and recommendations' and a link to section 8 was provided in sections 3 and 4.
- In section 3, the Table for sample size and input recommendations for the SQK-LSK114 kit was updated. Figures 1 and 2 were updated with data using R10.4.1 flow cells. Information on duplex basecalling was removed as it is no longer applicable. Links for new protocols were added.
- In section 5, 'RNA and cDNA sequencing kits', the Direct RNA Sequencing Kit XL (SQK-RNA004-XL) was included.
- In section 7, 'Expansion packs', the XL packs for the Sequencing Auxiliary Vials and Flow Cell Priming Kit were included. The following expansions were also added: Short Fragment Eliminator Kit, Ultra-Long Auxiliary Vials, and PCR Barcoding Expansion. Figure 4 for the Flow Cell Wash Kit was updated.
- In section 8, 'Sample input and recommendations', Figure 5 was updated.
- In section 9, 'Making the most of your flow cell', Flongle was removed from the Table as it is in Closed Store. More information was provided for the channel states panel and pore activity plots. The MinKNOW screenshots for pausing/stopping a run and starting a pore scan were updated.
- The section title 'Appendix' was changed to 'Sequences and reagents' for better visibility.
- In section 16, 'Kit reagents', a link to the sequence for the Lambda DNA control was provided.
- All links in the document were updated where applicable. All graphs were numbered and provided with a figure legend.
