- In 'Overview of the protocol', an important note on Kit14 sequencing and duplex basecalling info sheet was removed as this is a Legacy document and its information is no longer applicable.
- In 'Priming and loading the PromethION Flow Cell', the step to remove the flow cell from the fridge was moved to the top of this section and a step to complete a flow cell check was included.
- In 'Data acquisition and basecalling' information on duplex basecalling was removed as it is no longer applicable.
