Adaptive sampling is a software-based enrichment and depletion method unique to Oxford Nanopore Technologies that targets regions of interest during sequencing with no additional library preparation. By sequencing native DNA, modifications are preserved in targets and can be analysed alongside the nucleotide sequence.
The method is incorporated into MinKNOW™, the operating software that drives all nanopore sequencing devices. With real-time sequencing, it is possible for a sequence that represents a region of interest to be identified as the DNA strand passes through a nanopore. This is achieved through the rapid mapping of the beginning of the sequenced strand to the provided reference file.
