DNA methylation analysis of a tandem repeat specific to Coturnix japonica using nanopore sequencing

Elena Bolshakova is currently studying for a Master’s degree at the Herzen State Pedagogical University of Russia in Saint Petersburg with a major in Biology. Her work is dedicated to the study of the structural and functional organization of repetitive elements in the genome of the Japanese quail. She is also interested in exploring the possibilities of nanopore sequencing in the analysis of epigenetic modifications of DNA.


The role of tandem repeats is still poorly understood. In our study, we analysed the DNA methylation of the Coturnix japonica pericentromeric tandem repeat CjapSAT (MH475922). We have shown that most of the monomers are methylated, consistent with constitutive heterochromatin. Comparison of methylation patterns in DNA samples from blood cells and early embryos revealed differences in the degree of methylation of individual repeats. Nanopore sequencing allows us to identify the certain repeating elements of CjapSAT, localised on the particular chromosomes, and find some differences in the methylation of individual monomers in different regions of the genome.
